Acetyl Tetrapeptide-5

Skincare

Also known as: Eyeseryl, β-Ala-His-Ser-His, Carelab EYS

Limited Evidence

What is Acetyl Tetrapeptide-5?

A synthetic tetrapeptide developed specifically for the periorbital area, targeting under-eye puffiness and dark circles through reduction of capillary permeability and improvement of lymphatic drainage. Marketed under the trademark Eyeseryl by Lipotec (Lubrizol Advanced Materials).

How it works

Reduces edema formation in the under-eye area by strengthening blood vessel walls and reducing excessive capillary permeability — reducing plasma leakage that contributes to puffiness. Also inhibits glycation (protein cross-linking from glucose), which contributes to skin stiffening and loss of elasticity around the eye area. Some data suggests it supports lymphatic drainage by improving capillary vessel integrity.

What marketers claim

  • eliminates dark circles permanently
  • completely removes bags under the eyes
  • works as quickly as topical fillers

What evidence supports

  • manufacturer study (Lipotec): 20% reduction in under-eye edema measured by 3D imaging after 28 days at 4% concentration
  • in vitro data shows reduction in capillary permeability markers in endothelial cell cultures
  • anti-glycation activity documented in cell culture studies

Research evidence

Eyeseryl (Acetyl Tetrapeptide-5) for periorbital edema reduction

2009Clinical Trialn = 20 volunteers

Finding: 20 volunteers applied 4% Acetyl Tetrapeptide-5 eye cream twice daily for 28 days; 3D volumetric imaging showed ~20% reduction in periorbital volume vs baseline.

Limitation: Manufacturer-sponsored, no placebo control arm described, very small sample (20 subjects).

What to expect

Realistic timeline based on available research. Individual results vary.

Week 2–4

Under-eye puffiness may show mild improvement with consistent twice-daily application, particularly in the morning when edema is highest.

Month 2–3

Anti-glycation benefits and structural capillary integrity improvements require sustained use.

Safety notes & concerns

Full safety guide →
  • evidence limited to manufacturer-sponsored studies — no independent clinical trials
  • under-eye puffiness has multiple causes (genetics, fluid retention, allergies, sleep) — one peptide cannot address all of them
  • effective concentration in commercial products rarely disclosed or likely far below the studied 4%
  • dark circles from pigmentation or vascular visibility are different mechanisms from edema — this peptide specifically targets edema and permeability

Pairs well with

caffeine (complementary approach to under-eye puffiness)Argirelinevitamin K (for dark circles from broken capillaries)

Use caution with

fragrance and essential oils in the periorbital area (general caution for eye area products)

Frequently asked questions

Can Acetyl Tetrapeptide-5 remove dark circles?

It depends on the cause. Dark circles from puffiness (bags casting a shadow) may improve as edema reduces. Dark circles from visible blood vessels under thin skin may improve marginally via capillary permeability reduction. Dark circles from pigmentation (melanin deposits) are a different mechanism entirely and are not addressed by this peptide. Most people have more than one contributing factor.
